Cord blood hemopoietic progenitor cell toll like receptor expression and function: a mechanism underlying allergic inflammation in early life?

چکیده
Methods Freshly isolated, CD34-enriched human CB cells were stimulated with 10 μg/mL lipopolysaccharide (LPS) or 5 μM CpG ODN overnight. Flow cytometric analyses were used to evaluate surface and intracellular expression of TLR-2, TLR-4, TLR-9, as well as the hemopoietic cytokine receptors (HCR) IL-5R, IL-3R and GMCSFR; methylcellulose cultures were performed to assess CD34+ cell differentiation capacity into Eo/B CFU.

Silymarin inhibits Toll-like receptor 8 gene expression and apoptosis in Ramos cancer cell line
Objective: Silymarin is a herbal extract containing flavonolignans, and it has inhibitory effects against the growth of different cancer cell lines by inducing apoptosis. Toll-like receptors are suggested as a novel and attractive target to treat cancer. The current study aimed at examining the mechanism of silymarin-induced apoptosis in Ramos cells and investigating its effect...
متن کاملIL-4 and IL-13 regulate TLR expression and eosinophil-basophil differentiation of cord blood CD34+ progenitor cells
Background Intrauterine environmental exposures have been shown to influence neonatal immunity and subsequent allergic disease development [1]. We have previously shown that cord blood (CB) progenitor cells of high atopic risk infants have reduced toll-like receptor (TLR) expression and produce fewer lipopolysaccharide (LPS)-stimulated eosinophil-basophil (Eo/B) colonies, compared to lowatopic ...
متن کاملEvaluating the Effects of Cytomegalovirus Glycoprotein B on the Maturation and Function of Monocyte-derived dendritic cells
Background & Objectives: Interaction of cytomegalovirus glycoprotein B with toll-like receptors of dendritic cells leads to early signaling and innate immune responses. The aim of this study is to evaluate the effects of cytomegalovirus glycoprotein B on the maturation and function of monocyte-derived dendritic cells in treated groups in comparison with control groups. Materials & Methods: Bloo...
متن کامل3D study of capillary network derived from human cord blood mesenchymal stem cells and differentiated into endothelial cell with VEGFR2 protein expression
New blood forming vessels are produced by differentiation of mesodermal precursor cells to angioblasts that become endothelial cells (ECs) which in turn give rise to primitive capillary network. Human cord blood (HCB) contains large subsets of mononuclear cells (MNCs) that can be differentiated into endothelial-like cells in vitro. Human mononuclear progenitor cells were purified from fresh umb...
متن کاملMonocyte Expression of Toll-like Receptor-4 in Patients with Stable Angina Undergoing Percutanoeus Coronary Intervention
Background: Toll like receptors (TLRs) are well recognized players in inflammatory conditions. Among them TLR-4 is involved in chronic inflammatory processes such as formation of atherosclerotic plaques. Objective: The present study was aimed to examine the effects of percutanoeus coronary intervention (PCI) as a revascularization method on monocyte expression of hTLR-4 and on the serum levels ...
